MarPiM: Marine fungal biotech for early control of metastatic melanoma
General information
Malignant melanoma is an aggressive, early metastasising type of tumour. The treatment of metastatic malignant melanoma with conventional cancer therapeutics is currently only successful to a very limited extent: in more than 50 per cent of late-diagnosed melanoma patients, tumour cells have already migrated into the bone marrow. The cells that have spread from the primary tumour can migrate and cause occult metastases in the lungs, liver and brain as well as in the bone itself. The 5-year survival rate for metastases is only 14 per cent. There is therefore an immense clinical need for new active substances for the prevention of metastasising malignant melanoma. The preliminary work of the project partners has shown that extracts of a fungal symbiont derived from the native brown algae have a toxic effect against melanoma cell lines. The combination of innovative strategies from marine natural product chemistry, bioinformatics and machine learning-based metabolomics enables the rapid and targeted purification and chemical characterisation of marine fungal compounds with in vitro anti-melanoma activity. The aim of the MarPiM project is to optimise the biotechnological production of marine active substances in large quantities for preclinical testing. The planned work includes the production of sufficient substance quantities, the elucidation of the mode of action at the molecular level using innovative methods of miniaturised proteomics for the analysis of small cell numbers, as well as in vivo efficacy using modern imaging methods in animal models to determine clinical relevance.
